Family groupings based on Y-DNA marker values, using color to show differences between family groups.
Distances from the modal values are color-coded here.  See the legend below the chart.
For each individual and each marker tested, the number shown in the chart is the individual's value for that marker.  Closely related individuals will have almost identical marker values.  
BUT the problem is that unrelated people in the same haplogroup will ALSO share many marker values, so we have to be able to tell the difference between being related, vs. just being in the same haplogroup (or subgroup) but unrelated (within "genealogical time").
To do this, we 1) note the modal (~average) markers for the relevant haplogroup (or subgroup); 2) find the markers for each specific family that differ from the haplogroup modal values;  and 3) compare these unique markers when comparing different individuals.
A given family will have CHARACTERISTIC MARKERS that are different from their haplogroup's modal values.

In this chart, these family-specific markers are indicated by a vertical column where most members of a family have similar color shades and similar marker values. 
          For instance, descendants of Nathaniel Thrift of VA have markers 439, 460, YCAIIa, 576, CDYa etc that differ from their haplogroup's modal values. Most of these markers are in FTDNA's set of markers 26 through 37.
          If other people from the same haplogroup are tested, chances are they will be close to the haplogroup modal values, so they will probably have to test at least 37 markers, in order to compare these unique markers.
          However, the descendant of Isham Thrift has several unique markers (that differ from the haplogroup mode) even among the first 12 markers tested (390, 385a, 385b, 439).
          If other people from the same haplogroup are tested, they will only need to test 12 or 25 markers IF the goal is merely to see whether they are related to the known descendant of Isham Thrift.
          Clearly, Isham and Nathaniel have different markers, so they are unrelated.  Likewise, Isham Thrift's markers differ from those of the two Friths.
For the two descendants of Nathaniel Thrift, who are 6th cousins (once removed), the DNA results confirm the genealogical information: they ARE closely related but there are three differences (458, 464c, & 456), i.e. they are not extremely close.
This is actually quite reassuring.   If the DNA results disagreed with the known genealogy, we would have to investigate BOTH the genealogy and the DNA testing carefully to find the source of the discrepancy.  
More importantly, when the genealogy is unknown, the DNA results can show us whether or not two families are (probably) related.
Thus the DNA results tell us that despite the shared surname, Robert, Nathaniel and Isham Thrift are NOT related to each other. 
The two Friths appear to be unrelated (given the limited data we have for one of them so far).
Stay tuned, when I get time I will put up an analysis of non-surname matches.  For one line, this will be important information.

The R1b families above are displayed together to allow comparison with markers from the modal haplotype from haplogroup R1b1b2.  
However any family that is clearly a member of a SUBCLADE of R1b can be compared to the modal haplotype for their OWN subclade, for more specific results.
Jim Cullen's World Haplogroup & Haplo-I Subclade Predictor ( http://members.bex.net/jtcullen515/haplotest.htm ) predicts these are the most likely subclades for the indicated families:
          Nathaniel Thrift of Virginia:  R1b or R1b-North/South 2 (not a close match to either)
          Isham & William Thrift:  R1b
          John Frith? (& Realie Klinner) of Alabama:  R1b-Frisian (S21*) 
                    R1b-Frisian (S21*)  is a subclade of R1b1b2a1a1.    They are predicted to carry the S21 SNP marker but no known downstream SNP markers.
