Purpose of the project

 

The Strain Surname DNA Project was originally organized to determine whether two lines of American Strain families—that of Thomas Jefferson Strain and that of John Strain of Washington County, Tennessee—were related or not.   The project has since expanded to include other Strain families in America and abroad and recently received a sample from the Strayhorn family.  We hope to gather DNA samples from other families with similar surnames and determine whether these names are related and perhaps discover where these families originated.

Procedure

 

The original participants selected Family Tree DNA (FTDNA) of Houston, Texas to perform the testing.   Most of the participants to date have been given FTDNA’s 25-marker Y-chromosome DNA test, which tests 25 markers on the Y-chromosome.  Some participants have also taken the extended 37-marker test.  These tests examine specific areas on the Y-chromosome—which is found only in males—where there are identifiable repetitions of particular DNA sequences (Short Tandem Repeats, or STRs).  The number of repetitions is counted and is listed as the value for that marker.

 

While most chromosomes are shuffled around from one generation to the next, the Y-chromosome is passed with little alteration down the male line of descent, from father to son to grandson, etc.  So when males of the same surname are tested it is possible to determine whether they might have descended from a common male ancestor by comparing these selected markers on their Y-chromosome.

 

However, periodically mutations can occur which cause subtle changes in the Y-chromosome.  The enzyme responsible for “copying” DNA may add or leave out one or more repetitions of a particular DNA sequence, or environmental factors may cause a mutation.  Currently Family Tree DNA estimates that one mutation occurs, on average, about every 500 years or about 20 generations, although some markers are believe to mutate somewhat faster and others somewhat slower.  These mutations are passed on from father to son.  Though the mutations are infrequent they are unpredictable, and it is even possible for brothers to have slightly different Y-chromosomes. 

   

Because it is not possible to accurately predict when these mutations will occur it is impossible to accurately determine the exact relationship of two individuals based on their DNA results.  Their interrelatedness can only be stated as a probability.  But the more similar the results are to each other the more likely it is that the individuals shared a common male ancestor in the more recent past, while the more different they are the more likely it is that the common male ancestor was more remote in time.

Results

 

The following table shows the results of the Y chromosome DNA tests that have been processed so far by Family Tree DNA. 

 

We assume (1) that the selected markers are dependable indicators of common ancestry, (2) that the test results are valid and no samples were contaminated, switched or misread, and  (3) that all participants are actually descendants of the ancestors they claim.

 

Each column labeled DYS# refers to a particular marker on the Y chromosome.  The numbers in each column are the number of DNA sequence repetitions (STRs) found at that marker.  Family Tree has used these values to predict each individual’s Haplogroup, although it is necessary to test other slower-mutating snippets of DNA to confirm the Haplogroup.  Haplogroup is useful in determining where in the world an individual’s ancestors might have lived—for example, R1b indicates Western European origins.
